| Description: | YI-type shuttle integrative vector to incorporate a neoR cassette into human-derived (Alu-containing) YACs. Constructed by inserting a 1.2 kb 5'-EcoRI and BamHI-3' fragment containing S. cerevisiae HIS3 into the EcoRI and BamHI sites of pSV2-neo. A 0.3 kb BamHi fragment from BLUR13 was modified by adding an EcoRI site the SphI site and then cloned into the BamHI site between neoR and HIS3. If linearized with EcoRI and transformed into YAC-containing his3 deletion strains, the plasmid will target integration to Alu sequences and confer a His+ phenotype. The YAC can then be transferred to a eukaryotic cell by spheroplast fusion. The order of themajor features in this plasmid is: pMB1 ori - bla - HIS3 - BamHI - BLUR13.RI - BamHI - neoR. | ||
