| Description: | Plasmid YEp356 is a useful vector for studying control of gene expression in S. cerevisiae. The vector contains the E. coli lacZ gene, encoding β-galactosidase. The lacZ gene lacks the promoter, operator, transla-tion-initiation codon and the first eight codons. The plasmid harbors within the lacZ gene the polylinker region from M13mp18. DNA fragments containing a promoter and translation-initiation region can be inserted into these sites, to form fusion proteins, if the reading frames are properly aligned. The formation of β-galactosidase fusion proteins in S. cerevisiae can be detected on solidified medium agar plates by addition to the medium of the chromogenic substrate X-gal. The vector can replicate autonomously in S. cerevisiae due to the presence of the 2μ yeast plasmid replicon, and in E. coli. It contains selection markers for S. cerevisiae (URA3) and for E. coli (amp, pyrF). | ||
