| Description: | Plasmid pTRP56 serves as the starting material for preparation of the vector primer. This vector primer can be prepared by digestion with BamHI and KpnI. The BamHI site provides the cohesive end at which the vector primer is joined to the linker fragment. The 3' protruding end of the KpnI site can be used for tailing with T residues which can form basepairs with the polyA tract of the mRNA. Close to the KpnI site the primer fragment also contains the polyadenylation signal from the TRP5 gene of S. cerevisiae. Plasmid pTRP56 has the ability to replicate in S. cerevisiae due to the presence of an element (ARS) allowing autonomous replication in S. cerevisiae. The vector contains an origin of replication for E. coli and selection markers for S. cerevisiae (TRP1) and for E. coli (amp, trpC) | ||
