Description:
|
This plasmid confers resistance to G418 in yeast. An episomal shuttle vector for subcloning and recovery of DNA from S. cerevisiae. pNF2 can be used to create genomic libraries by inserting DNA from Sau3AI partial digests into the BamHI site. The inserts can be excised readily by the SalI sites that flank the BamHI site. pNF2 was constructed from pNF1 by inserting a BamHI linker into the filled-in SalI site. pNF1 was constructred from pYE24 by inserting a KanR-containing fragment from pMC874 into the BamHI site. The vector comprises a 2.2 kb EcoRI fragment containing the origin of replication of the B-form of the 2 micron yeast plasmid, a 1.1 kb HindIII fragment from S. cerevisiae containing the URA3 gene, a 2.6 kb Sau3A fragment containing the kan gene from Tn903 inserted into the EcoRI, HindIII and BamHI site, respectively, of pBR322. The orientation of the kan gene is not given.
