Description:

The phasmid can be used as cloning vectors for S. cerevisiae and E. coli. It is derived from pEMBL9 by insertion of yeast sequences and can be rapidly switched in E. coli between the plasmid and phage modes of replication. It contains the ampicillin resistance gene and the lacZalpha gene for selection and detection, respectively, of recombinant plasmid in E. coli. It can be propagated in S. cerevisiae because of the presence of the 2 micron plasmid replicon, and in E. coli. The phasmid differs in their cloning sites and in its genetic marker which is used for selection of transformation in S. cerevisiae. The vector pEMBL Ye23 contains the yeast URA3 gene which can be used as a selective marker in S. cerevisiae and E. coli. Phasmid pEMBL Ye23 comprises a 3.1 kb HindIII fragment from Yep24 containing the URA3 gene and the 2 micron yeast phasmid replicon, inserted into the ClaI site of pEMBL9.
