The plasmid is used for the expression of Cpf1 and NgAgo in B. subtilis，and can replicate in E. coli.
To build plasmid pHT‐XCR6, plasmid pHT‐XC was first constructed by adding the F. novicida U112 Cpf1 (Zetsche
et al., 2015) controlled by the xylose‐induced promoter PxylA into the E. coli‐B. subtilis shuttle vector pHT01. Then plasmid pHT‐XCR6 was obtained by inserting NgAgo (Fu et al., 2019) controlled by the isopropyl‐beta‐D‐thiogalactopyranoside (IPTG) induced promoter Pgrac100 into plasmid pHT‐XC. NgAgo was modified (preserving
650–887 aa, D663A, D738A) to abolish its potential enzyme activity.
